Research Interests
Based on the liquid chromatography-mass spectrometry, we focus on the development of novel methods for quantitative analysis of the proteomes in biological samples, such as cell lines and tissues.
Stable isotopic labeling of proteins/peptides through chemical reactions has become widely used in quantitative proteomics studies, as this strategy minimizes the technical variability in sample preparation and MS analysis. However, chemical labeling often causes undesired side reactions, compromising the analytical sensitivity and dynamic range. We attempt to investigate the chemical mechanism of the side reactions and develop novel labeling methods allowing specific and efficient labeling of proteins/peptides.
Histones play a crucial role in the epigenetic regulation via its various post-translational modifications (PTMs). However, deciphering histone PTMs, especially their combinatorial patterns, by LC-MS remains an area of active development. Based on the histone derivatization method we established previously,,we attempt to establish novel analytical method for accurate and robust characterization of histone PTMs on both bottom-up and middle-up approaches.
