We all need sleep, but why do we need sleep? This is an important and fundamental biological question. Dr. Dong focuses on the research of the mechanism of sleep-wake regulation, mainly dissecting the neural circuits that control of sleep and wakefulness and monitoring the of neuromodulator dynamic during sleep-wake cycles. By utilizing optogenetics, chemogenetics, viral tracing, and in vivo neuronal activity recording techniques, he has deciphered the neural circuits that regulate sleep and wakefulness in the striatum (Current Biology, 2022; eLife, 2017) and in the dorsal part of the deep mesencephalic nucleus (Cell Discovery, 2022). Furthermore, leveraging novel genetically encoded neurotransmitter/neuromodulator sensors (GENIs), he has elucidated the dynamic release pattern of histamine during sleep-wake cycles (Neuron, 2023). Dr. Dong is using GENIs to study dynamic changes of neuromodulatorsincluding monoamines, neuropeptides and lipids, during sleep-wake cycles, focusing on those with unique sleep-related dynamic patterns to uncover their biological significance.
