癲癇是一種重要的神經(jīng)疾病,人群發(fā)病率約為1%。我國(guó)癲癇患者總數(shù)近1000萬(wàn),其中,耐藥性癲癇(drug-resistant epilepsy, DRE)患者約占30%,尚無(wú)有效的臨床藥物。課題組采用在體腦電記錄,免疫磁珠細(xì)胞分選,雙光子鈣成像,光纖鈣信號(hào)記錄和全細(xì)胞膜片鉗等神經(jīng)科學(xué)領(lǐng)域前沿技術(shù)探究癲癇等重大腦疾病的發(fā)生發(fā)展機(jī)制,我們重點(diǎn)關(guān)注顳葉性癲癇和腦中風(fēng)伴發(fā)癲癇癥狀中膠質(zhì)細(xì)胞與神經(jīng)元、免疫細(xì)胞與膠質(zhì)細(xì)胞互作的機(jī)制(Nat Neurosci, 2023;Nat Neurosci, 2025;Neuron, in revision;Cell, in revision)。具體包括但不限于以下四方面研究?jī)?nèi)容:
1) 研究顳葉性癲癇疾病中膠質(zhì)細(xì)胞和神經(jīng)元互作及其與癲癇進(jìn)展之間的關(guān)系,以此為突破口尋找抗癲癇治療的全新靶點(diǎn);
2) 開展癲癇腦(顳葉性癲癇和腦中風(fēng)伴發(fā)的癲癇等)等重大腦疾病中外周免疫細(xì)胞和中樞神經(jīng)細(xì)胞(膠質(zhì)細(xì)胞)互作的機(jī)制研究;
3) 外界環(huán)境刺激會(huì)誘導(dǎo)癲癇發(fā)生,其機(jī)制尚未明了。我們重點(diǎn)關(guān)注聲音和光等外界環(huán)境刺激誘導(dǎo)癲癇發(fā)作的神經(jīng)環(huán)路機(jī)制。
4) 癲癇常伴發(fā)有認(rèn)知和情緒障礙,其共病機(jī)制尚不清楚。我們擬開展癲癇共患認(rèn)知障礙及情感障礙的神經(jīng)機(jī)制研究。
